运动经PKC-Nrf2途径调节肝HO-1表达及抗氧化能力的研究The Regulation Role of Exercise on Heme Oxygenase-1 Expression and Antioxidant Capacity in the Liver by PKC-Nrf2 Pathway

摘要(Abstract):
为探讨规律耐力运动通过PKC-Nrf2途径提高大鼠肝血红素加氧酶-1(HO-1)表达及抗氧化能力(T-AOC)的效应,将24只成年雄性SD大鼠随机分为安静对照组(A)、运动组(B)与白屈菜赤碱运动组(C)。B、C组进行8周较大强度的持续耐力跑,但C组在每次运动前进行PKC抑制剂即白屈菜赤碱(CHE)处理。实验中观察各组肝细胞膜PKC及肝细胞总PKC的活性水平,Western Blotting观察肝细胞核Nrf2及肝细胞HO-1的蛋白表达水平,以及观察肝细胞HO-1活性和T-AOC水平,结果发现:与A组相比,B组肝细胞膜PKC及肝细胞总PKC的活性显著升高、细胞膜PKC活性/肝细胞总PKC活性的值显著升高(P<0.05),细胞核中的Nrf2蛋白含量(核转位)显著增加(P<0.05),肝细胞HO-1的蛋白表达水平、活性水平及肝细胞T-AOC等均显著升高(P<0.05);耐力运动引起B组中的上述变化在C组中被明显遏制。综上可见,较大强度的规律耐力运动可经过PKC-Nrf2途径,诱导肝细胞HO-1蛋白表达而提高肝脏抗氧化能力。
